Yes, it is possible to update multiple sites with your plugins. However, not from custom repository specifically, but ZIP files can be uploaded from your local storage to MainWP Dashboard and then installed to child sites.

Alternatively, you can also try our Favorites extension, but keep in mind that you will have to manually upload new plugin versions once they become available to the Favorites extension: Favorites - MainWP WordPress Management
